The production of magnesium protoporphyrin monomethyl ester by Rhodopseudomonas spheroides.

چکیده
A monoester of magnesium protoporphyrin has been isolated by Granick (1961) from a chlorophyll-less mutant of Chlorella and from etiolated barley leaves treated with 8-aminolaevulic acid and aaoc-bipyridyl. It was considered likely that this material was a methyl ester that Granick then proposed as an intermediate in the biosynthesis of chlorophyll. The extraction from Rhodop8eudomona spheroides of an enzyme system which methylates magnesium protoporphyrin to form the monomethyl ester (Tait & Gibson, 1961) has been given as evidence for the participation of this metalloporphyrin in bacteriochlorophyll biosynthesis. In this paper is described the detection and identification of magnesium protoporphyrin monomethyl ester, which was secreted into the medium by intact cells of R. spheroide8, growing on complete medium and carrying out normal bacteriochlorophyll synthesis.

The Inhibition of Bacteriochlorophyll Biosynthesis in Rhodopseudomonas Spheroides by 8-hydroxyquinoline.
Granick (1948, 1950, 1961) has isolated different mutants of Chlorella which are unable to synthesize chorophyll and which accumulate magnesium protoporphyrin, magnesium vinyl phaeoporphyrin a5 and a monoester of magnesium protoporphyrin, presumed to be the monomethyl ester. Granick (1961) has proposed that these compounds are intermediates in chlorophyll biosynthesis, and it has been suggested...
متن کاملThe accumulation of bacteriochlorophyll precursors by mutant and wild-type strains of Rhodopseudomonas spheroides.
1. Two mutant strains of Rhodopseudomonas spheroides, which are blocked in the synthesis of bacteriochlorophyll, accumulate pigments. These have been tentatively identified as magnesium 2,4-divinylphaeoporphyrin a(5) monomethyl ester and the magnesium derivative of 2-devinyl-2-hydroxyethyl-phaeophorbid a, formed by mutant 2/73 and 2/21 respectively. 2. Maximum extracellular production of these ...
متن کاملControl of magnesium-protoporphyrin chelatase activity in Rhodopseudomonas spheroides. Role of light, oxygen, and electron and energy transfer.
1. Magnesium-protoporphyrin chelatase activity, previously shown in whole cells of Rhodopseudomonas spheroides, could not be demonstrated in cell-free extracts prepared in different ways, although spheroplasts retained moderate activity. Slight activity was detected also in whole cells of Rhodospirillum rubrum. 2. The effects on the activity of the enzyme of inhibitors of electron and energy tr...
متن کاملProtochlorophyll-like Pigment Produced by Rhodopseudomonas spheroides
The identification of intermediates in the biosynthesis of chlorophyll has been largely dependent upon the use ofmutants of Chlorella that are unable to synthesize chlorophyll but accumulate in the medium various tetrapyrrole compounds assumed to be precursors (for a review see Granick & Mauzerall, 1961). In particular, Granick (1950) identified magnesium vinylphaeoporphyrin a5 monomethyl ester...
متن کاملMutant strains of Rhodopseudomonas spheroides lacking delta-aminolevulinate synthase: growth, heme, and bacteriochlorophyll synthesis.
Two mutant strains of Rhodopseudomonas spheroides were described which lacked delta-aminolevulinate synthase activity. They required delta-aminolevulinate for growth; they did not respond to protoporphyrin or magnesium photoporphyrin, and only poorly to hemin. Synthesis of cytochromes and heme by mutant H-4 was dependent upon delta-aminolevulinate; this strain did not form bacteriochlorophyll e...
